Myxothiazol

Description

Inhibitor of CIII (inhibits also CI; G. Lenaz). Myxothiazol binds to the Qo site of CIII (very close to cytochrome bL) and inhibits the transfer of electrons from reduced QH2 to the Rieske Iron sulfur protein [1].

Abbreviation: Myx

Reference: MiPNet09.12

MitoPedia topics: Inhibitor

HRR: Chemicals; Media"Chemicals; Media" is not in the list (Oxygraph-2k, TIP2k, O2k-Fluorometer, pH, NO, TPP, Ca, O2k-Spectrophotometer, O2k-Manual, O2k-Protocol, ...) of allowed values for the "Instrument and method" property.

Application in HRR

Myx: Myxothiazol (Myxothiazol from Myxobacterium myxococcus fulvus, ~95%, HPLC), C25H33N3O3S2); Sigma T5580, 1 mg, store at 4 °C: FW = 487.7

Caution: Very toxic. Chemical is again available from Sigma with the product number cited above 2011-04-18. The old product number was M 5779 . Alternatively use Antimycin A as a Complex III inhibitor.

Preparation of 5 mM stock solution (dissolved in 99.9% Ethanol):

1) weigh 1 mg of Myxothiazol into a small glass vial
2) add 2.05 ml Ethanol
3) divide into 0.2 ml (use glass vials)
4) store at -20 °C

Oxygraph-2k Manual Titrations: MiPNet09.12

  • Titration volume: 1 µl using a 10 µl syringe
  • Final conc. in 2 ml O2k-chamber: 0.5 µM
