Description
Octanoylcarnitine is a medium-chain fatty acid (octanoic acid: eight-carbon saturated fatty acid) covalently linked to carnitine.
Abbreviation: Oct
Reference: pdf
MitoPedia topics: Substrate and metabolite
Carnitine O-octanoyltransferase is an enzyme that catalyzes the chemical reaction:
- octanoyl-CoA + L-carnitine ↔ CoA + L-octanoylcarnitine
Thus, the two substrates of this enzyme are octanoyl-CoA and L-carnitine, whereas its two products are CoA and L-octanoylcarnitine.
Application in HRR
Oct: Octanoylcarnitine (DL-Octanoyl-carnitine-HCL, C15H30NO4Cl); TOCRIS Bioscience, Cat. No. 0605, 50 mg, store at R.T, desiccate); FW = 323.85.
Alternative: (±)-Octanoylcarnitine chloride, BioTrend, Cat. No. BS0136, 50 mg, store desiccate at RT; FW = 323.86.
Caution: Chemicals stored in the fridge or freezer should be allowed to reach room temperature before opening.
Preparation of 100 mM stock solution (dissolved in H2O):
- Weigh 32.4 mg of octanoylcarnitine and dissolve in 1 ml H2O.
- Divide into 0.2 ml portions.
- Store at -20 °C.
Oxygraph-2k manual titrations into 2 ml O2k-chamber
- Titration volume: 4 µl using a 10 µl syringe.
- Final concentration: 0.2 mM in MiR05 or MiR06. Optimum Oct concentration has to be evaluated, particularly in media with BSA concentrations different from MiR06.
