DatLab-Analysis templates

Description

Go in DatLab to Mark statistics (F2), select which type of marks you want to export ("All marks in plot" or "DL-protocol marks", with 3 possibilities each), then click on [Copy to clipboard] to copy selected values and paste them to a DatLab-Analysis template for numerical and graphical data analysis.

DatLab-Analysis templates for O2k-analysis of respiratory flux - strategy

DatLab-Analysis templates are made available for specific SUIT protocols, to express respiration as oxygen flux or oxygen flow normalized for sample concentration, and as flux control ratios, FCR and flux control factors, FCF.

Colour codes

  • Yellow: you have to copy/enter something here.
  • Cyan: a hyperlink to another cell within the DatLab-Analysis template.
  • Light blue: a hyperlink to a web page.

Guidelines

  1. Make sure that the DatLab-Analysis template corresponds with the protocol of your experiment. Open the chosen template and click at tab #1. Check if the reference mark names (row 33) correspond to the protocol of your experiment.
  2. Select the tab "Navigation". Fill in the sample number (Sample.Subsample: i.e. 1.1) in column E of your experiment and create a hyperlink to each individual DatLab file in column F. It is recommended to save the DatLab-Analysis template and the DatLab files in the same subdirectory.
  3. Set the [[mark]s on the flux (or flow, slope) plot of your subsample. Press [F2] to open Mark statistics (O2 slope neg.) and [Copy to clipboard].
  4. In the DatLab-Analysis template, select the tab # (#1; #2; ...) for your sample (tab #1 may be different from the Sample number in your experiment [F3]). Click on the hyperlink [↓ Subsample entry #a; #b; #c; ..] and [β†’ Mark statistics], taking you to the yellow cell in column AA. Press [Ctrl-V] to paste the data of your subsample from Mark statistics in DatLab into the yellow cell under column AA.
  5. Copy the graph in DatLab. Go to the DatLab-Analysis template, click on [← Graph], and paste into the yellow cell for the subsample. Adjust the length of the graph to 20 cm (8 inch).
  6. Proceed with all subsamples (the template provides 8 subsamples: #a,#b,#c,#d,#e,#f,#g and #h for each sample).
  7. Repeat for all samples. Currently 6 tabs are provided for independent samples (#1,#2,#3,#4,#5 and #6).
  8. Select tab Cohort. Enter a short acronym for your cohort in cell E1 (yellow). This tab provides automatically a summary of your experimental cohort.


Concept

  1. The normalization for sample concentration is transferred into a DatLab-Analysis template, since (1) in some cases the final sample concentration is not known until a later time point (e.g. protein or CS determination), and different markers may be used for different aspects of the analysis; (2) the sample (isolated mitochondria, homogenate, cell suspensions) is diluted during titrations, and the dilution effect is calculated in the DatLab-Analysis template.
  2. A logical next step: instrumental O2 background parameters may not be known at the time of the experiment, particularly when brackting instrumental quality control is performed (before and after the experiment). Finally optimized background parameters may be calculated at the end of a complete experimental series, combining several relevant instrumental O2 background tests. Up to now, the DatLab-Analysis files do not document the instrumental O2 background parameters. For quality control each individual DatLab file has to be checked. Therefore, the logical next step is to export from Mark statistic only slopes (O2 slope neg.) for complete data analysis in the DatLab-Analysis template. There, the instrumental O2 background correction is easily implemented, adjustments of parameters are quickly performed, actual parameters used can be much more quickly checked, and documentation for tracing is provided in the DatLab-Analysis files in a more complete and compact form.

  1. Select a Layout for DatLab graphs with the plot of O2 flux or O2 flow normalized for the sample concentration.
  2. Set marks on the plot for O2 flux or O2 flow.
  3. Press [F2] (or click on Marks \ Statistics). Then click on [Copy to clipboard] to copy selected values and paste them to an DatLab-Analysis template for flux analysis.
